Revenge counter should do Blackstar in, at least from what I remember.

It most likely would, however most of the damage BS deals is piercing/cutting. I think Mel could only take so many of that type of damage compared to blunt damage.

His stamina and endurance is crazy though, does Blackstar have what it takes to put him down?

I think so. Talk about crazy stamina, Black Star had his spine broken and received many broken bones, open wounds and even swallowed an explosion before spitting it out. His physical strength is also up there.

If they're equal at base then wouldn't a transformed Mel take the win? He gets a pretty big boost when he turns.

Strength alone won't give him the win though. Meliodas can take a beating for a while. It's actually part of the way he fights.

Black Star arguably can take more than Mel can. The punishment he takes is crazy not the least of which being having his spine broken and not stopping.
